Magnesium Supplementation Attenuates Pulmonary Hypertension via Regulation of Magnesium Transporters

چکیده

Pulmonary hypertension (PH) is characterized by profound vascular remodeling and altered Ca 2+ homeostasis in pulmonary arterial smooth muscle cells (PASMCs). Magnesium ion (Mg ), a natural antagonist cofactor for numerous enzymes, crucial regulating diverse cellular functions, but its roles PH remains unclear. Here, we examined the of Mg transporters development. Chronic hypoxia monocrotaline induced significant adult male rats. It was associated with reduction [Mg ] i PASMCs, increase gene expressions Cnnm2 , Hip14 Hip14l Magt1 Mmgt1 Mrs2 Nipa1 Nipa2 Slc41a1 Slc41a2 Trpm7 ; upregulation SLC41A1, SLC41A2, CNNM2, TRPM7 proteins; downregulation SLC41A3 mRNA protein. supplement attenuated pressure, right heart hypertrophy, medial wall thickening arteries, reversed changes expression transporters. Incubation PASMCs high concentration markedly inhibited PASMC proliferation migration, increased apoptosis, whereas low level produced opposite effects. siRNA targeting Slc41a1/2, Cnnm2, promoted apoptosis; Slc41a3 overexpression also caused similar Moreover, or incubation hypoxia-induced nuclear translocation NFATc3 PASMCs. The results, first time, provide supportive evidence that participate development modulating proliferation, supplementation attenuates through regulation involving signaling pathway.

Magnesium transporters: properties, regulation and structure.

The chemistry of Mg2+ is unique amongst biological cations, and the properties of Mg2+ transport systems reflect this chemistry. Prokaryotes carry three classes of Mg2+ transport systems: CorA, MgtA/B and MgtE. CorA and MgtE are widely distributed in both Eubacteria and Archaea, while the MgtA/B class is found primarily in the Eubacteria. Eukaryotic homologs of CorA, although clearly functional...

Vascular biology of magnesium and its transporters in hypertension.

Magnesium may influence blood pressure by modulating vascular tone and structure through its effects on myriad biochemical reactions that control vascular contraction/dilation, growth/apoptosis, differentiation and inflammation. Magnesium acts as a calcium channel antagonist, it stimulates production of vasodilator prostacyclins and nitric oxide and it alters vascular responses to vasoconstrict...

Magnesium supplementation prevents the development of alcohol-induced hypertension.

The effect of chronic alcohol administration on blood pressure was investigated in 7-week-old Wistar rats. Tail-cuff blood pressure was significantly higher in rats who received 15% ethanol in drinking water than in control rats. Intracellular free calcium concentration of lymphocytes was increased, while magnesium concentration of erythrocyte, aorta, and skeletal muscle and erythrocyte ouabain...

Oral magnesium supplementation in patients with essential hypertension.

To elucidate the effects of magnesium on high blood pressure, a 4-week study of oral magnesium supplementation (MgO 1 g/day) was conducted in 21 outpatients with uncomplicated essential hypertension. During the study, blood pressure and intraerythrocyte sodium concentration decreased significantly, and the erythrocyte ouabain-sensitive 22Na efflux rate constant (Kos) and intraerythrocyte magnes...

Lowered magnesium in hypertension.

Lowered Magnesium in Hypertension To the Editor: We read with interest the article by Joosten et al1 dealing with urinary magnesium excretion and risk of hypertension—the PREVEND (Prevention of Renal and Vascular End-Stage Disease) study. The authors examined 5511 participants. A total of 1172 developed hypertension. The urinary magnesium excretion was associated with risk of hypertension in an...
